Bramich, Narelle J., and Helen M. Cousins. Effects of sympathetic nerve stimulation on membrane potential, [Ca]i, and force in the toad sinus venosus. Am. J. Physiol. 276 (Heart Circ. Physiol. 45): H115–H128, 1999.—The effects of sympathetic nerve stimulation on beat rate, force, intracellular Ca21 concentration ([Ca]i) measured using fura 2, and membrane potential were recorded from the spontaneously beating toad sinus venosus. Short trains of stimuli evoked an increase in the beat rate and force. During this tachycardia the amplitude of pacemaker action potentials was not changed, but there was an increase in the basal level of [Ca]i with little change in peak [Ca]i measured during each action potential. Depletion of intracellular Ca21 stores with caffeine (3 mM) abolished all responses to sympathetic nerve stimulation. The effects of caffeine were fully reversible. Caffeine (3 mM), in the presence of the Ca21-ATPase inhibitor thapsigargin (30 μM), abolished irreversibly the chronotropic and inotropic responses evoked by sympathetic nerve stimulation. Ryanodine (10 μM) attenuated, but did not abolish, these responses. These results suggest that, in the toad sinus venosus, increases in force and beat rate evoked by sympathetic nerve stimulation result from the release of Ca21 from intracellular Ca21 stores.

متن کاملParallel metabotropic pathways in the heart of the toad, Bufo marinus.
This study examined the transduction pathways activated by epinephrine in the pacemaker region of the toad heart. Recordings of membrane potential, force, and intracellular Ca(2+) concentration ([Ca(2+)](i)) were made from arrested toad sinus venosus. Sympathetic nerve stimulation activated non-alpha-, non-beta-adrenoceptors to evoke a membrane depolarization and a transient increase in [Ca(2+)...
متن کاملVagal and Sympathetic Effects on the Pacemaker Fibers in the Sinus Venosus of the Heart* by Otto
The response of excitable cells to inhibitory nerve impulses has already been studied with intracellular electrodes in several preparations. In the spinal cord of the cat an inhibitory volley sets up a transient polarization of the motoneuron, preventing it from discharging (Brock, Coombs, and Eccles (4)). In the crustacean nerve muscle junction inhibitory impulses have a dual effect. They atte...
متن کاملVagal and Sympathetic Effects on the Pacemaker Fibers in the Sinus Venosus of the Heart
1. Action potentials from sinus venosus and auricle fibers of spontaneously beating frog hearts have been recorded with intracellular electrodes. 2. Sinus fibers show a slow depolarization, the pacemaker potential, during diastole. The amplitude of this potential varies in different parts of the sinus. In some fibers the membrane potential falls by 11 to 15 mv. during diastole and the transitio...
متن کاملProtective effects of an interaction between vagus nerve and melatonin on gastric ischemia/reperfusion: the role of oxidative stress
Objectives:Vagal pathways in gastrointestinal tract are the most important pathways that regulate ischemia/reperfusion (I/R). Gastrointestinal tract is one of the important sources of melatonin production. The aim of this study was to investigate probable protective effect of the interaction between vagus nerve and melatonin after I/R. Materials and methods:This study was performed in male rat...
